This groundbreaking work unveils a paradigm-shifting platform for in situ protein assembly and precision purification, merging split intein engineering with microbial consortia design. Pioneering a ""one-pot"" biosynthesis strategy, the book details how engineered Escherichia coli equipped with the ePop autolysis system synchronously releases intein-tagged protein fragments, while Pichia pastoris secretes complementary domains via optimized eukaryotic secretion pathways. By integrating quorum-sensing controls and split intein-mediated trans-splicing, this system achieves extracellular protein reconstitution directly in culture, bypassing costly in vitro purification steps . The platform's modularity enables logic-gated synthesis of multi-domain proteins, exemplified by its applications in antibiotic resistance engineering and context-responsive biomaterials. Central to this innovation is the rational engineering of the Mycobacterium xenopi GyrA intein. Through targeted mutagenesis (Cys1Ala, Ser179Gly, His197Gln), we developed a pH/calcium-responsive self-cleaving tag that releases bioactive human basic fibroblast growth factor (bFGF) with >85% efficiency. The Ser179Gly mutant exhibits rapid C-terminal cleavage under alkaline conditions (50 mM Tris-HCl, pH 8.5), while the Cys1Ala/His197Gln double mutant boosts precursor yield 3.2-fold-a breakthrough for high-titer production. A novel 3-step purification cascade (cation exchange → heparin affinity → gel filtration) recovers 85% of bioactive bFGF from lysates, validated by LC-MS/MS and MTT assays. Parallel protocols for human epidermal growth factor (hEGF) and growth hormone (hGH) demonstrate platform versatility.

This book provides a systematic and comprehensive interdisciplinary overview of ductile mode cutting of brittle materials, covering a range of topics from the fundamental physics to engineering practices. Discussing the machining mechanics and material properties, it explains the fundamental mechanism of ductile-to-brittle transition in the cutting of brittle materials. It also presents theoretical modeling and molecular dynamic simulation to demonstrate that ductile mode cutting can be achieved under certain conditions, as well as extensive experimental studies that produced smooth and damage-free surfaces on different materials, such as silicon, glass, tungsten carbide and calcium fluoride. Lastly, it explores how the ductile mode cutting performance and machinability of brittle materials can be further improved by hybrid machining processes like ultrasonic vibration and thermal-assisted cutting technologies in order to meet industry demands.
Water-resisting Property and Key Technologies of Grouting Reconstruction of the Upper Ordovician Limestone in North China’s Coalfields
This book examines the water resistance capacity of the Upper Ordovician limestone and its feasibility as a water barrier to achieve safe and green mining. Mine water inrush events often occur during coal mine construction and production; they account for a large proportion of the coal mine disasters and accidents in China, second only to gas explosions. As mining depths and mining intensity continue to increase, the hydrogeological conditions encountered are becoming more complex. This book describes in-situ methods designed to test the water resistance of the limestone layer, as well as specific grouting techniques developed to transform this layer into a barrier that can prevent water inrush during mining. The innovative technologies, which were applied and validated in two coal mines, are applicable to other coal mines or any underground engineering works.

International Teaching Assistants' Professional Identity Development
Informed by Critical Theory and Poststructuralist Theory and the intersections of agency, power, ideology, discourse, capital, and language, this study investigates how three ITAs construct their professional identities as instructors at a U. S. university. To gain an in-depth understanding of ITAs' professional identities development, the researcher uses a qualitative approach with a multi-case study design to examine various data and variables including a) undergraduates' feedback to ITAs' instruction, b) ITAs' English language use in academic settings and its influence on their teaching, and c) the role of ITAs' supervisors on their professional practice. Through narrative analysis, the researcher analyzes data from interviews, classroom observations, and research journals. Findings suggest that both course evaluations and ITAs' interactions with supervisors and colleagues influence their professional identity formation. Additionally, the ITAs' English language use in academic settings reflects their desire of becoming native English speakers for an audience of mostly US undergraduates.

Materials Characterisation and Mechanism of Micro-Cutting in Ultra-Precision Diamond Turning

Materials Characterisation and Mechanism of Micro-Cutting in Ultra-Precision Diamond Turning

Springer-Verlag Berlin and Heidelberg GmbH Co. K
2018
nidottu
This book presents an in-depth study and elucidation on the mechanisms of the micro-cutting process, with particular emphasis and a novel viewpoint on materials characterization and its influences on ultra-precision machining. Ultra-precision single point diamond turning is a key technology in the manufacture of mechanical, optical and opto-electronics components with a surface roughness of a few nanometers and form accuracy in the sub-micrometric range. In the context of subtractive manufacturing, ultra-precision diamond turning is based on the pillars of materials science, machine tools, modeling and simulation technologies, etc., making the study of such machining processes intrinsically interdisciplinary. However, in contrast to the substantial advances that have been achieved in machine design, laser metrology and control systems, relatively little research has been conducted on the material behavior and its effects on surface finish, such as the material anisotropy of crystalline materials. The feature of the significantly reduced depth of cut on the order of a few micrometers or less, which is much smaller than the average grain size of work-piece materials, unavoidably means that conventional metal cutting theories can only be of limited value in the investigation of the mechanisms at work in micro-cutting processes in ultra-precision diamond turning.
From Mathematics to Philosophy (Routledge Revivals)
First published in 1974. Despite the tendency of contemporary analytic philosophy to put logic and mathematics at a central position, the author argues it failed to appreciate or account for their rich content. Through discussions of such mathematical concepts as number, the continuum, set, proof and mechanical procedure, the author provides an introduction to the philosophy of mathematics and an internal criticism of the then current academic philosophy. The material presented is also an illustration of a new, more general method of approach called substantial factualism which the author asserts allows for the development of a more comprehensive philosophical position by not trivialising or distorting substantial facts of human knowledge.

This book provides different financial models based on options to predict underlying asset price and design the risk hedging strategies. Authors of the book have made theoretical innovation to these models to enable the models to be applicable to real market. The book also introduces risk management and hedging strategies based on different criterions. These strategies provide practical guide for real option trading. This book studies the classical stochastic volatility and deterministic volatility models. For the former, the classical Heston model is integrated with volatility term structure. The correlation of Heston model is considered to be variable. For the latter, the local volatility model is improved from experience of financial practice. The improved local volatility surface is then used for price forecasting. VaR and CVaR are employed as standard criterions for risk management. The options trading strategies are also designed combining different types of options and they have been proven to be profitable in real market. This book is a combination of theory and practice. Users will find the applications of these financial models in real market to be effective and efficient.

A Logical Journey

A Logical Journey

Hao Wang (1921-1995) was one of the few confidants of the great mathematician and logician Kurt Goedel. A Logical Journey is a continuation of Wang's Reflections on Goedel and also elaborates on discussions contained in From Mathematics to Philosophy. A decade in preparation, it contains important and unfamiliar insights into Goedel's views on a wide range of issues, from Platonism and the nature of logic, to minds and machines, the existence of God, and positivism and phenomenology. The impact of Goedel's theorem on twentieth-century thought is on par with that of Einstein's theory of relativity, Heisenberg's uncertainty principle, or Keynesian economics. These previously unpublished intimate and informal conversations, however, bring to light and amplify Goedel's other major contributions to logic and philosophy. They reveal that there is much more in Goedel's philosophy of mathematics than is commonly believed, and more in his philosophy than his philosophy of mathematics. Wang writes that "it is even possible that his quite informal and loosely structured conversations with me, which I am freely using in this book, will turn out to be the fullest existing expression of the diverse components of his inadequately articulated general philosophy." The first two chapters are devoted to Goedel's life and mental development. In the chapters that follow, Wang illustrates the quest for overarching solutions and grand unifications of knowledge and action in Goedel's written speculations on God and an afterlife. He gives the background and a chronological summary of the conversations, considers Goedel's comments on philosophies and philosophers (his support of Husserl's phenomenology and his digressions on Kant and Wittgenstein), and his attempt to demonstrate the superiority of the mind's power over brains and machines. Three chapters are tied together by what Wang perceives to be Goedel's governing ideal of philosophy: an exact theory in which mathematics and Newtonian physics serve as a model for philosophy or metaphysics. Finally, in an epilog Wang sketches his own approach to philosophy in contrast to his interpretation of Goedel's outlook.

Today, privacy is one of the most hotly debated topics worldwide. The book aims to balance the development of personal rights in a country that has historically valued collective rights over those of the individual. The protection of privacy is not an issue that has been emphasised during the rapid development of economic laws in China. However, the accompanying development of greater government-based regulation of these laws’ implementation has led to greater invasions of personal privacy. This study attempts to provide a way forward for China to address the ever-increasing concerns about the protection of privacy and puts forward a legislative model for protection. This is achieved after a thorough analysis of the threats to privacy protection in China, a critical evaluation of the level of current privacy protection in China, and an analysis of the privacy laws in a series of developed nations based on common law and civil law.
