CPX loaded Nanostructured lipid carrier (NLC) were successfully developed as a potential topical delivery system for the treatment of wounds. The drug and its potential excipients for NLC were screened by preformulation studies viz. organoleptic evaluation, melting point analysis, UV-Visible spectra, solubility studies, partition coefficient analysis, Fourier transform-Infrared (FT-IR) analysis and Differential Scanning calorimetric (DSC) analysis. After successful characterization of drug, formulation development was carried out for the selection and optimization of solid lipid, liquid lipid, solid lipid to liquid lipid ratio, surfactant and co-surfactant concentration, formulation technique, process parameters viz. PDI, Sonication time and EE%. The characterizations of the prepared NLC formulations were assessed by means of shape, particle size distribution, zeta potential analysis, drug entrapment efficiency, and in vitro drug release studies to select the optimized NLC formulation. The permeation study revealed that the permeability parameters were significantly higher for NLC-based gel formulation as compared to marketed formulation.

Poor water solubility of active pharmaceutical ingredients (APIs) presents a significant challenge in oral drug administration, leading to reduced bioavailability and compromised therapeutic effectiveness. This study aimed to address this issue by developing an optimized itraconazole nanosuspension for intranasal delivery, potentially enhancing drug availability across the blood-brain barrier. Then a nanosuspension was prepared and characterized using various analytical techniques. FTIR and DSC analyses verified the drug's chemical structure and crystalline state . A Box-Behnken Design was employed to optimize the formulation, resulting in nanoparticles with favorable size, polydispersity index, zeta potential, and entrapment efficiency. TEM imaging revealed a core-shell structure of the nanoparticles. The optimized nanosuspension demonstrated improved drug release compared to a conventional suspension. These findings suggest that the developed itraconazole nanosuspension has potential to enhance the drug's solubility and bioavailability.

Lymphatic filariasis, considered globally as a neglected tropical disease (NTD), is a parasitic disease caused by microscopic, thread-like worms, the parasite damages the lymph system. Most of the infected people have no symptoms and will never develop clinical symptoms. Fixed dose combination kits have emerged as a potential technique for the treatment of lymphatic filariasis. LF is treated with a mix of pharmacological therapy, vector control, and preventative measures. FDC kits for the treatment of LF are made up of two or more antifilarial medicines that target distinct phases of the parasite's life cycle. These kits make treatment regimens easier to follow, increase patient compliance, and improve overall treatment results. The use of FDC kits in LF therapy has various advantages. It removes the need for patients to take various pills or capsules at different times, decreasing therapy complexity and boosting adherence. Antifilarial medicine combination and dose standardisation decreases the possibility of incorrect drug selection or dosage mistakes. Furthermore, the combination of various medications in FDC kits may aid in the prevention of drug resistance.

Gastro-retentive schwimmende Verabreichungssysteme f r Medikamente haben sich als ein effizientes Mittel zur Verbesserung der Bioverf gbarkeit und kontrollierten Verabreichung vieler Medikamente erwiesen. Es wurden verschiedene Techniken f r die gastro-retentive, ortsspezifische Verabreichung von Medikamenten berichtet. Alle diese Techniken haben jedoch ihre eigenen Vor- und Nachteile. Zu den Vorteilen eines schwimmenden Verabreichungssystems geh ren: Erh hung der Verweildauer im Magen aufgrund des Auftriebs, ortsspezifische Verabreichung von Arzneimitteln in den Magen, kontrollierte Freisetzung von Arzneimitteln ber einen l ngeren Zeitraum, geringere Dosierungsh ufigkeit, gezielte Therapie von lokalen Beschwerden im oberen GIT-Bereich, bessere therapeutische Wirkung von Arzneimitteln mit kurzer Halbwertszeit, erh hte Absorption von im Magen l slichen Arzneimitteln und geringere Schwankungen der Arzneimittelkonzentration.

La absorci n de medicamentos en el tracto gastrointestinal es un procedimiento muy variable y la prolongaci n de la retenci n g strica de la forma de dosificaci n prolonga el tiempo de absorci n. El sistema de administraci n de drogas flotantes gastro-retentivas ha surgido como un medio eficiente de mejorar la biodisponibilidad y la administraci n controlada de muchas drogas. Se ha informado de varias t cnicas para la administraci n de drogas en sitios espec ficos de retenci n g strica. Sin embargo, todas estas t cnicas tienen sus propias ventajas y desventajas. Las ventajas del sistema de administraci n flotante de drogas incluyen: el tiempo de retenci n g strica se incrementa debido a la flotabilidad, se puede obtener la administraci n de drogas en un sitio espec fico del est mago, la liberaci n controlada de drogas por un per odo prolongado, la disminuci n de la frecuencia de las dosis, la terapia dirigida a dolencias locales en la parte superior del GIT, un mejor efecto terap utico de las drogas de vida media corta, el aumento de la absorci n de drogas solubles en el est mago y la reducci n de las fluctuaciones de la concentraci n de drogas.
