Gloriosa superba L. est une importante esp ce de plante m dicinale appartenant la famille des Colchicaceae. La m thode conventionnelle de propagation de cette plante est celle des tubercules, qui est lente et reste dormante sous terre pendant les mois secs (septembre-mars). Sa production est saisonni re et elle est sensible de nombreux ravageurs. La mauvaise propagation et la surexploitation par la population locale et les entreprises pharmaceutiques sont les principaux facteurs responsables de la diminution de la taille de la population. C'est pourquoi le livre rouge de l'UICN l'a class e parmi les plantes menac es d'extinction. Il est donc urgent de conserver cette plante. La micropropagation est donc une m thode alternative et efficace pour surmonter tous les probl mes. On pense que les r sultats de ces tudes seront utiles pour d velopper un protocole appropri pour la conservation et l'utilisation durable de cette plante.

La Gloriosa superba L. un'importante specie di pianta medicinale appartenente alla famiglia delle Colchicaceae. Il metodo convenzionale di propagazione di questa pianta attraverso i tuberi, che sono lenti e rimangono dormienti sottoterra durante i mesi secchi (settembre-marzo). La sua produzione stagionale ed soggetta a molti parassiti. La scarsa propagazione e l'eccessivo sfruttamento da parte della popolazione locale e delle aziende farmaceutiche sono i principali fattori responsabili della diminuzione della sua popolazione. Per questo motivo, stata dichiarata pianta a rischio di estinzione dal Libro Rosso della IUCN. quindi urgente conservare questa pianta. Pertanto, la micropropagazione un metodo alternativo ed efficace per superare tutti i problemi. Si ritiene che i risultati di questi studi saranno utili per sviluppare un protocollo adeguato per la conservazione e l'uso sostenibile della pianta.

Gloriosa superba L. ist eine wichtige Heilpflanze, die zur Familie der Colchicaceae geh rt. Die konventionelle Methode der Vermehrung dieser Pflanze erfolgt durch Knollen, die langsam sind und w hrend der trockenen Monate (September bis M rz) unter der Erde ruhen. Ihre Produktion ist saisonal und sie ist anf llig f r viele Sch dlinge. Die schlechte Vermehrung in Verbindung mit der berm igen Ausbeutung durch die rtliche Bev lkerung und die Pharmaunternehmen sind die Hauptfaktoren f r den R ckgang der Population. Daher wurde sie vom Roten Buch der IUCN als gef hrdete Pflanze eingestuft. Es ist daher dringend notwendig, diese Pflanze zu erhalten. Daher ist die Mikrovermehrung eine alternative und wirksame Methode zur berwindung aller Probleme. Man geht davon aus, dass die Ergebnisse solcher Studien bei der Entwicklung eines geeigneten Protokolls f r die Erhaltung und nachhaltige Nutzung der Pflanze hilfreich sein werden.

Gloriosa superba L. is an important medicinal plant species belonging to the family Colchicaceae. It is a perennial tuberous climbing herb having an attractive wavy-edged orange-red flower. The conventional method of propagation of this plant is through tubers which is slow and remains dormant underground during dry months (September- March). Its production is seasonal having susceptibility toward many pests. The poor propagation coupled with over-exploitation by the local population as well as pharmaceutical companies are the main factors responsible for its diminishing population size. So, it has been affirmed as an endangered plant by the IUCN Red Data Book. Hence, there is an urgent need to conserve this plant. Therefore, micropropagation is an alternate and effective method to overcome all the problems. It is believed, that the findings of such studies would help develop suitable protocols for conservation and its sustainable use.

The cells were harvested from bacterial culture isolate by centrifugation and were suspended in Tris-HCl buffer of pH 8.0. The optical density (O. D.) of cells was measured at 600nm and each time cells of 21.5 optical densities (O. D.) were disrupted and used for the purification of the enzyme. Purification of superoxide dismutase from SPB-13. Ammonium sulphate precipitation method was used for concentrating the proteins. After this, the enzyme was further purified by DEAE-Sepharose column. The DEAE-sepharose column was loaded with 3ml of concentrated protein. The enzyme activity was only recorded in fractions ranging from 22, 23, 24, 25, 26, 27 and 28. These fractions were polled together. The polled fractions along with crude enzyme were checked for purity by SDS-PAGE. The maximum activity of superoxide dismutase of SPB-13 was found to be in Tris-HCl buffer, 60mM molarity, pH 8.0, incubation time was found to be 2.0 minutes, incubation temperature 450C. An enzyme concentration of 10 g was found to be optimum. Thermostability of superoxide dismutase was checked and enzyme was found to be thermostable upto 550C for 60 minutes.
